Arythrex " Celecoxib "

Composition: Each capsule contains: Celecoxib 100 or 200mg.

Properties: Arythrex® (Celecoxib) is a non-steroidal anti-inflammatory drug that exhibits anti-inflammatory, analgesic and anti-pyretic activities. These effects are due to inhibition of prostaglandin synthesis, primarily via inhibition of Cyclooxygenase-2 (COX-2).  At therapeutic concentrations in humans, Arythrex® does not inhibit the cyclooxygenase-1 (COX-1) isoenzyme.

Pharmacokinetics:
-    Arythrex® (Celecoxib) is well absorbed from the gastro-intestinal tract and peak plasma concentrations were attained within 3 hours after an oral dose.
-    Administration of Arythrex® with a high-fat meal resulted in a 1-2 hours delay in the peak plasma concentration levels and a 10-20% increase in the bioavailability.
-    Arythrex® Capsules can be administered without regard to the timing of meals.
-    At therapeutic plasma concentrations, Arythrex® is about 97% bound to plasma proteins principally albumin and, to a lesser extent, ?1- acid glycoprotein.
-    The plasma elimination half-life is about 11 hours. Celecoxib is metabolized in the liver to inactive metabolites principally by the Cytochrome P-450 isoenzyme 2C9.
-    Approximately 57% and 27% of a single oral dose of Celecoxib is eliminated in the feces and urine respectively. Less than 3% is excreted in the feces and urine unchanged. The primary metabolite is Carboxylic Acid metabolite (73% of the dose).
Indications: Arythrex® is indicated for the relief of pain and inflammation in:
-    Osteoarthritis
-    Rheumatoid arthritis
-    Gouty arthritis
-    Non-articular disorders such as: Myositis, tendinitis,  tenosynovitis and bursitis
-    Acute pain of any origin
-    Dysmenorrhea
Contra-Indictions: =    Known hypersensitivity to Celecoxib.
=    Arythrex® should not be given to patients who have experienced allergic-type reactions to sulfonamides.
=    NSAIDs, including Celecoxib, generally are contra-indicated in patients in whom asthma, urticaria or other sensitivity reactions are precipitated by aspirin or other NSAIDs.
Adverse Effects:-    Gastro-intestinal disturbances.
-    Allergic reactions.
-    Aggravated hypertension.
-    Headache.
-    Dizziness
-    Rarely adverse renal, ocular and otic effects.
Dosage & Administration: The dosage of Arythrex® should be adjusted according to individual requirements and response using the lowest possible effective dosage.
-    Osteoarthritis: For the symptomatic treatment of osteoarthritis, the usual adult dose is 200mg per day administered as a single dose or as 100mg twice daily.
-    Rheumatoid arthritis: The usual adult dose is 100-200mg twice daily.
-    Pain and dysmenorrhea
-    For the relief of acute pain or dysmenorrhea, the usual initial adult dose is 400mg given as a single dose followed by an additional dose of 200mg, if necessary, on the first day. For continued relief, 200mg may be administered twice daily as needed.
